Genetic diversity of Kalopanax septemlobus (Thunb.) Koidz. at the northern edge of the range according to chloroplast DNA sequencing data

An analysis of nucleotide polymorphism of intergenic spacers psbA–trnH, rpl32–trnL, and trnS–trnfM of chloroplast DNA in accessions of Kalopanax eptemlobus at the northern end of the range (Russian Far East) was carried out. The genetic diversity of the total sample from the south of Primorsky krai was very low (h = 0.142, π = 0.0001), and the genetic diversity of the total sample from Sakhalin Island was medium (h = 0.600, π = 0.0018). Analysis of molecular variance showed that about 86% of all genetic variability is accounted for by differences between the two geographic regions (ΦST = 0.857, P < 0.0001). Nucleotide substitutions and variations of indels revealed three haplotypes (H1–H3), of which the H3 haplotype was found only on Sakhalin Island. The haplotype H1 is widespread in Primorsky krai, whose frequency was 92.4%. The observed variability of K. septemlobus in the Russian Far East reflects by history of its biogeography.
