Roholtiella mixta sp. nov. (Nostocales, Cyanobacteria): morphology, molecular phylogeny and carotenoid content

A Calothrix-like cyanobacterial strain was isolated from a sample collected from the entrance zone of Shumyaschiy Ponor cave (Chelyabinsk Region, South Urals, Russia). This isolate showed diagnostic features of the recently described genus, Roholtiella, i.e. chains of arthrospores and thalli not tapering to a hair. Molecular analyses based on 16S rRNA gene, 16S–23S internal transcribed spacer (ITS) sequences, and spacer secondary structure revealed that our isolate was a distinct lineage within the genus clade that we describe as Roholtiella mixta sp. nov. High-performance liquid chromatography identified ?-carotene, zeaxanthin, 3-hydroxyechinenone (4-keto-3-hydroxy-?-carotene), echinenone, caloxanthin, ?-cryptoxanthinoleate, and antheraxanthin in R. mixta. Antheraxanthin was recorded for the second time only in cyanobacteria species.
